Ao mesmo tempo, nunca se mostrou t\u00e3o necess\u00e1ria. Como unidade cient\u00edfica do Complexo, o Instituto de Pesquisa Pel\u00e9 Pequeno Pr\u00edncipe precisou equilibrar-se sobre a fina linha entre a preven\u00e7\u00e3o por meio de distanciamento social e o engajamento na maior batalha dos cientistas em muitas d\u00e9cadas.[\/vc_column_text][vc_column_text css_animation=&#8221;fadeIn&#8221;]A pandemia teve tr\u00eas principais efeitos nos trabalhos do Instituto:<\/p>\n<ul>\n<li>em raz\u00e3o das exig\u00eancias de isolamento social, reduziu ou mesmo paralisou o acompanhamento de participantes em alguns dos projetos ent\u00e3o em andamento;<\/li>\n<li>diminuiu encontros presenciais entre os pesquisadores. Em tempos normais, o Instituto chega a receber cem alunos por dia \u2013 entre gradua\u00e7\u00e3o, mestrado, doutorado e p\u00f3s-doutorado. Com a pandemia, foram dez por turno (20 por dia). O atraso no desenvolvimento de certas pesquisas tornou-se inevit\u00e1vel;<\/li>\n<li>deflagrou uma s\u00e9rie de pesquisas relacionadas \u00e0 COVID-19 (foram 17 estudos ao todo).<\/li>\n<\/ul>\n<p>A produ\u00e7\u00e3o de artigos (com e sem colabora\u00e7\u00e3o de alunos) aumentou: a maioria dos pesquisadores publicou mais do que nos anos anteriores.<\/p>\n<p>A capacidade de r\u00e1pida adapta\u00e7\u00e3o da equipe e os esfor\u00e7os em conhecer melhor a doen\u00e7a que parou boa parte do planeta evidenciam o compromisso do Instituto com a pesquisa na \u00e1rea da sa\u00fade e sua contribui\u00e7\u00e3o para transformar o futuro \u2013 usando a ci\u00eancia para levar melhorias de vida a todos.[\/vc_column_text][\/vc_column][vc_column column_padding=&#8221;no-extra-padding&#8221; Com uma \u00fanica gota de sangue do animal, o exame detecta se ele foi vacinado ou est\u00e1 com o v\u00edrus;<\/p>\n<p>*<strong> Processo com nanotecnologia<\/strong>: o processo de an\u00e1lise pode ser usado para testes em humanos e animais. Ainda em 2020, foi adaptado para detectar o SARS-CoV-2 em pessoas. Esse estudo remete a um dos grandes feitos do Instituto de Pesquisa Pel\u00e9 Pequeno Pr\u00edncipe: a descoberta, tempos atr\u00e1s, de uma muta\u00e7\u00e3o gen\u00e9tica heredit\u00e1ria (<em>TP53<\/em>-R337H) que pode causar o <a href=\"http:\/\/www.pelepequenoprincipe.org.br\/pesquisas-resultados-dna.php\" target=\"_blank\" rel=\"noopener\">tumor de c\u00f3rtex adrenal<\/a>, um c\u00e2ncer que, apesar de raro, tem incid\u00eancia maior na Regi\u00e3o Sul do Brasil. O achado motivou a inclus\u00e3o do <a href=\"http:\/\/pequenoprincipe.org.br\/noticia\/teste-do-pezinho-pode-diagnosticar-cancer-e-imunodeficiencias-primarias\/\" target=\"_blank\" rel=\"noopener\">diagn\u00f3stico dessa muta\u00e7\u00e3o no Teste do Pezinho<\/a> no Paran\u00e1. Em 2020, foi publicado um desdobramento desse trabalho, agora em parceria com o St. Jude Children\u2019s Research Hospital, dos Estados Unidos, e o Hospital de Amor (Barretos). Em cerca de 70% dos casos existe tamb\u00e9m uma defici\u00eancia imunol\u00f3gica associada. Este projeto contribui para elabora\u00e7\u00e3o de estrat\u00e9gias de preven\u00e7\u00e3o de infec\u00e7\u00e3o nos p\u00f3s-operat\u00f3rio, evitando per\u00edodos prolongados de interna\u00e7\u00e3o e at\u00e9 mesmo \u00f3bito.[\/vc_column_text][vc_column_text css_animation=&#8221;fadeIn&#8221;]<strong>Imaginologia, prote\u00e7\u00e3o radiol\u00f3gica e radioterapia<\/strong><\/p>\n<p><em>Tema: Radiografia em UTIs neonatais<br \/>\n<\/em><em>Objetivo<\/em>: propor um n\u00edvel de refer\u00eancia de dose m\u00ednima com qualidade (NRD) para esses exames. Eles s\u00e3o extremamente importantes para diagnosticar e acompanhar pacientes de UTI neonatal, mas beb\u00eas e crian\u00e7as s\u00e3o mais sens\u00edveis \u00e0 radia\u00e7\u00e3o: devem receber a menor dose necess\u00e1ria que gere uma imagem de qualidade para os m\u00e9dicos. O estudo recorre a dos\u00edmetros termoluminescentes a fim de avaliar a dose de radia\u00e7\u00e3o recebida por partes do corpo e encontrar a quantidade m\u00ednima necess\u00e1ria para cada tipo de exame.<\/p>\n<p><em>Tema: Modelagem computacional e radioterapia<br \/>\n<\/em><em>Objetivo:<\/em> buscar radioterapias mais exatas e din\u00e2micas. A modelagem computacional tem sido usada para apoiar as pesquisas cient\u00edficas e a rotina cl\u00ednica.<\/p>\n<p><em>Tema: M\u00e9todos de imobiliza\u00e7\u00e3o em radiocirurgia intracraniana<br \/>\n<\/em><em>Objetivo: <\/em>verificar qual posi\u00e7\u00e3o traz melhores resultados. Foram selecionados 50 casos de pacientes submetidos ao procedimento. Os desvios de posicionamento est\u00e3o sendo identificados por um sistema computacional.[\/vc_column_text][vc_column_text]<strong>Medicina molecular e bioinform\u00e1tica<\/strong><\/p>\n<p><em>Tema: Desenvolvimento de tumores<br \/>\n<\/em><em>Objetivo<\/em>: prever o risco de desenvolvimento de tumores. Algumas partes do genoma humano podem ter um n\u00famero de c\u00f3pias de genes normais por c\u00e9lula diferente de dois, e essa varia\u00e7\u00e3o ajuda a prever o risco de desenvolvimento de tumores. O projeto busca criar uma ferramenta computacional mais eficiente do que as atuais para identificar o n\u00famero de c\u00f3pias por c\u00e9lula.<\/p>\n<p><em>Tema: Albinismo oculocut\u00e2neo<\/em><\/p>\n<p><em>Objetivo:<\/em> o albinismo oculocut\u00e2neo \u00e9 uma doen\u00e7a gen\u00e9tica, rara, que causa altera\u00e7\u00f5es de pigmenta\u00e7\u00e3o de pele e cabelo, e altera\u00e7\u00f5es oftalmol\u00f3gicas. Apenas 25% dos pacientes desempenham de forma independente as atividades di\u00e1rias, devido \u00e0s altera\u00e7\u00f5es visuais. O projeto, que contou com a colabora\u00e7\u00e3o das equipes da Oftalmologia e Dermatologia do Hospital Pequeno Pr\u00edncipe e pesquisadores do Instituto Pel\u00e9 Pequeno Pr\u00edncipe e do Icahn School of Medicine at Mount Sinai, dos Estados Unidos, trabalhou n\u00e3o apenas os aspectos cl\u00ednicos, mas tamb\u00e9m moleculares do albinismo oculocut\u00e2neo. Os resultados da pesquisa j\u00e1 foram publicados, e foram descritas novas variantes gen\u00e9ticas associadas ao albinismo. Genes do albinismo est\u00e3o relacionados com processos de pigmenta\u00e7\u00e3o da pele (melanog\u00eanes), e isso pode trazer mais conhecimento sobre a fisiopatologia do c\u00e2ncer de pele.<\/p>\n<p><em>Tema: Bioinform\u00e1tica<\/em><\/p>\n<p><em>Objetivo:<\/em> a bioinform\u00e1tica \u00e9 essencial para as an\u00e1lises de dados gerados a partir do sequenciamento do DNA, do RNA e do microbioma, por exemplo. No Instituto de Pesquisa, diversos grupos trabalham com a bioinform\u00e1tica e, al\u00e9m de utilizarem softwares dispon\u00edveis no mercado, tamb\u00e9m trabalham com o aprimoramento e desenvolvimento de algoritmos e softwares. Um dos projetos desenvolve um software para an\u00e1lise de \u201cvariantes de n\u00famero de c\u00f3pias\u201d em dados de sequenciamento de exoma, com o objetivo de identificar \u00e1reas de perdas ou ganhos na sequ\u00eancia de DNA, que s\u00e3o importantes tanto para caracteriza\u00e7\u00e3o de doen\u00e7as raras quando na fisiopatologia do c\u00e2ncer.[\/vc_column_text][vc_column_text]<strong>Microbiologia e doen\u00e7as infecciosas<\/strong><\/p>\n<p><em>Tema: Microbiota respirat\u00f3ria<br \/>\n<\/em><em>Objetivo: <\/em>analisar a microbiota (conjunto de microrganismos que colonizam ou infectam pessoas) de pacientes com fibrose c\u00edstica, verificando eventuais rela\u00e7\u00f5es entre a diversidade e abund\u00e2ncia dos diversos grupos bacterianos e a evolu\u00e7\u00e3o cl\u00ednica dos pacientes.<\/p>\n<p><em>Tema: Genoma de fungos<br \/>\n<\/em><em>Objetivo<\/em>: sequenciar o genoma completo dos fungos <em>Candida haemulonii<\/em>, identificando os genes resistentes aos antif\u00fangicos e os respons\u00e1veis pela maior incid\u00eancia da doen\u00e7a. Com base nas caracter\u00edsticas gen\u00e9ticas espec\u00edficas, os profissionais de sa\u00fade podem escolher os melhores tratamentos.<\/p>\n<p><em>Tema: Leucemia linfobl\u00e1stica aguda (LLA)<\/em><\/p>\n<p><em>Objetivo: <\/em>avaliar as altera\u00e7\u00f5es que o tratamento quimioter\u00e1pico em crian\u00e7as e adolescentes com leucemia linfobl\u00e1stica aguda (LLA) pode acarretar nos padr\u00f5es de express\u00e3o g\u00eanica e na microbiota ent\u00e9rica (conjunto de micro-organismos que colonizam o intestino). Al\u00e9m disso, esses dados s\u00e3o tamb\u00e9m analisados no contexto de altera\u00e7\u00f5es gen\u00e9ticas espec\u00edficas da LLA, tamb\u00e9m pesquisadas nesse projeto. Financiado pelo Programa Nacional de Apoio \u00e0 Aten\u00e7\u00e3o Oncol\u00f3gica (Pronon), com esse projeto os pesquisadores querem compreender as associa\u00e7\u00f5es entre as mudan\u00e7as que ocorrem nas c\u00e9lulas da leucemia e na microbiota intestinal ao longo do tratamento, al\u00e9m de entender quais s\u00e3o os fatores-chave de sobrevida, quais as respostas terap\u00eauticas e a susceptibilidade a infec\u00e7\u00f5es.[\/vc_column_text][vc_column_text]<strong>Neuroci\u00eancias<\/strong><\/p>\n<p><em>Tema: Esclerose tuberosa<br \/>\n<\/em><em>Objetivo<\/em>: conhecer com mais detalhes os aspectos neurol\u00f3gicos e as possibilidades de diagn\u00f3stico e tratamento dessa doen\u00e7a gen\u00e9tica que pode acometer qualquer \u00f3rg\u00e3o pela ocorr\u00eancia de tumores. Os achados da pesquisa ressaltaram a import\u00e2ncia do diagn\u00f3stico precoce \u2013 e tamb\u00e9m de interven\u00e7\u00f5es capazes de reduzir a morbidade e o risco de complica\u00e7\u00f5es futuras e, assim, ter efeitos positivos na qualidade de vida dos pacientes e de suas fam\u00edlias.<\/p>\n<p><em>Tema: S\u00edndrome de opsoclonus-mioclonus (SOM)<br \/>\n<\/em><em>Objetivo<\/em>: conhecer mais sobre essa enfermidade rara, uma desordem neuroinflamat\u00f3ria devastadora quando n\u00e3o diagnosticada nem tratada precocemente. Existe apenas um estudo publicado por brasileiros, e n\u00e3o h\u00e1 protocolo para o tratamento da doen\u00e7a. A pesquisa do Instituto avalia o perfil neuropsicol\u00f3gico e neurol\u00f3gico de pacientes com o diagn\u00f3stico de SOM acompanhados no Ambulat\u00f3rio de Doen\u00e7as Raras, do Servi\u00e7o de Neurologia do Hospital Pequeno Pr\u00edncipe, e busca estabelecer um protocolo espec\u00edfico.<\/p>\n<p><em>Tema: Autismo<\/em><\/p>\n<p><em>Objetivo: <\/em>o transtorno do espectro autista (TEA) tem como um dos principais desafios o diagn\u00f3stico precoce. Um dos estudos do Instituto identificou fatores de risco para o desenvolvimento de TEA em beb\u00eas que passaram por UTI neonatal, o que pode contribuir para o desenvolvimento de estrat\u00e9gias de interven\u00e7\u00e3o, diagn\u00f3stico precoce e melhor progn\u00f3stico para essas crian\u00e7as.<\/p>\n<p>Al\u00e9m disso, outro estudo avaliou aspectos do ambiente familiar de escolares com TEA, indicando que a maioria das fam\u00edlias apresenta problemas de relacionamento, com perdas sociais. Foi observado alto n\u00edvel estresse nas fam\u00edlias de crian\u00e7as com TEA e uma grande preocupa\u00e7\u00e3o com o futuro dessas crian\u00e7as. Esses dados podem contribuir para auxiliar um melhor planejamento e melhorar a qualidade de vida das fam\u00edlias e crian\u00e7as com TEA.[\/vc_column_text][vc_column_text]<strong>Terapia celular e farmacol\u00f3gica<\/strong><\/p>\n<p><em>Tema: Esclerose m\u00faltipla<\/em><\/p>\n<p><em>Objetivo: <\/em>avaliar efeitos da terapia celular na encefalomielite autoimune experimental. Dois tipos de terapia celular est\u00e3o sendo avaliadas: terapia com c\u00e9lulas-tronco mesenquimais oriundas de cord\u00e3o umbilical humano e terapia com c\u00e9lulas precursoras neuronais. Esse trabalho traz mais dados pr\u00e9-cl\u00ednicos sobre o uso da terapia celular no tratamento de esclerose m\u00faltipla, podendo, futuramente, ser realizado estudo cl\u00ednico que venha a contribuir na melhoria da qualidade de vida dos pacientes com a doen\u00e7a.<\/p>\n<p><em>Tema: Terapias neurodegenerativas<\/em><\/p>\n<p><em>Objetivo:<\/em> caracterizar os RNAs n\u00e3o codificantes presentes nas ves\u00edculas extracelulares a partir de c\u00e9lulas-tronco mesenquimais para desenvolver biomarcadores e novas terapias, tendo como perspectivas futuras aplica\u00e7\u00f5es tanto em estudos pr\u00e9-cl\u00ednicos como em cl\u00ednicos com foco em doen\u00e7as neurodegenerativas e do neurodesenvolvimento.<\/p>\n<p><em>Tema: Doen\u00e7a de Parkinson<\/em><\/p>\n<p><em>Objetivo:<\/em> as c\u00e9lulas-tronco t\u00eam capacidade regenerativa e potencial de diferencia\u00e7\u00e3o. Evid\u00eancias sugerem que o efeito terap\u00eautico dessas c\u00e9lulas adv\u00e9m de produtos extracelulares, como os microRNAs. Diante disso, o estudo prop\u00f5e uma terapia baseada em microRNA. Os microRNAs que n\u00e3o apresentam efeitos t\u00f3xicos s\u00e3o incorporados em nanoemuls\u00f5es e utilizados em testes pr\u00e9-cl\u00ednicos. A doen\u00e7a de Parkinson \u00e9 induzida em ratos, e os animais s\u00e3o tratados por quatro a oito semanas com o nanomedicamento. Assim, espera-se o desenvolvimento de um nanomedicamento seguro para uma proposi\u00e7\u00e3o futura de estudo cl\u00ednico. A pesquisa foi aprovada no Programa Nacional de Gen\u00f4mica e Sa\u00fade de Precis\u00e3o, lan\u00e7ado pelo governo federal para incentivar o desenvolvimento cient\u00edfico e tecnol\u00f3gico nas \u00e1reas de gen\u00f4mica e sa\u00fade de precis\u00e3o no \u00e2mbito do SUS, al\u00e9m de impulsionar o desenvolvimento da ind\u00fastria gen\u00f4mica nacional.<\/p>\n<p>Ainda sobre a doen\u00e7a de Parkinson, outro estudo busca o desenvolvimento de uma nova formula\u00e7\u00e3o para medicamentos utilizados no tratamento, principalmente por meio de nanotecnol\u00f3gicos que possam aumentar a efic\u00e1cia da medica\u00e7\u00e3o, mas com uma concentra\u00e7\u00e3o menor, reduzindo assim os efeitos colaterais.<\/p>\n<p><em>Tema: Doen\u00e7as neurol\u00f3gicas<\/em><\/p>\n<p><em>Objetivo: <\/em>caracterizar as c\u00e9lulas precursoras neuronais diferenciadas a partir de c\u00e9lulas-tronco mesenquimais do tecido adiposo. A aplica\u00e7\u00e3o dessas c\u00e9lulas requer uma melhor compreens\u00e3o sobre os mecanismos respons\u00e1veis pela diferencia\u00e7\u00e3o, al\u00e9m da import\u00e2ncia de suas caracteriza\u00e7\u00f5es para padroniza\u00e7\u00e3o de protocolos e valida\u00e7\u00e3o para a sua futura utiliza\u00e7\u00e3o no tratamento de doen\u00e7as neurol\u00f3gicas.<\/p>\n<p><em>Tema: Tratamento de colite ulcerativa<br \/>\n<\/em><em>Objetivo: <\/em>investigar os efeitos de fibras extra\u00eddas da casca do maracuj\u00e1 e da gabiroba-verde (<em>Campomanesia adamantium<\/em>) no tratamento da colite ulcerativa. Essa \u00e9 uma doen\u00e7a inflamat\u00f3ria intestinal cr\u00f4nica, caracterizada por inflama\u00e7\u00f5es recorrentes, diarreia, dor abdominal e perda da funcionalidade do intestino. Em crian\u00e7as, tende a ser mais grave do que em adultos.<\/p>\n<p><em>Tema: Tratamento de mucosite intestinal<br \/>\n<\/em><em>Objetivo<\/em>: verificar os efeitos terap\u00eauticos das fibras do baga\u00e7o da guavira, subproduto da polpa da fruta, e de polissacar\u00eddeos do maracuj\u00e1-amarelo contra essa enfermidade \u2013 que costuma aparecer como rea\u00e7\u00e3o ao tratamento de diversos tipos de c\u00e2ncer, causando les\u00f5es no intestino. Lidar com esse problema significa dar um conforto maior aos pacientes oncol\u00f3gicos.<\/p>\n<p><em>Tema: Exposi\u00e7\u00e3o a agrot\u00f3xicos<br \/>\n<\/em><em>Objetivo<\/em>: avaliar uma s\u00e9rie de respostas biol\u00f3gicas a dois grupos de agrot\u00f3xicos muito utilizados no Brasil: dicamba e glifosato. Uma delas \u00e9 sobre o soro terap\u00eautico contra o novo coronav\u00edrus, desenvolvido a partir do material biol\u00f3gico de cavalos. A expectativa \u00e9 iniciar outros cerca de 50 projetos em 2021, muitos deles com potencial terap\u00eautico.
